The Aurora Biolabs KRAS G12V Nucleotide Exchange Assay Kit is a TR-FRET based assay designed to detect GTP loading on KRAS G12V in the presence of SOS1, a guanine nucleotide exchange factor. This kit uses Tag2-labeled KRAS protein and a terbium-labeled anti-Tag2 antibody (HTRF donor). When KRAS binds to fluorescently labeled GTP (HTRF acceptor), the donor and acceptor come into close proximity. Upon excitation at 340nm, energy transfer occurs, and emission at 665nm is measured. This assay quantitatively detects active GTP-bound KRAS G12V, ideal for inhibitor screening. Each kit supports 384 reactions.
